
One of the first peptides I ever experimented with was SS-31, an impressive mitochondrial repair peptide that I wrote about in this post. But it wasn’t long after that I discovered MOTS-c, an injectable peptide that works differently yet synergistically with SS-31. While SS-31 repairs damaged mitochondria, MOTS-c helps them function more efficiently and adapt to stress.
Although I didn’t experience dramatic changes with MOTS-c, as you might with energy-boosting supplements or performance enhancers, I still consider it a foundational part of my longevity and cellular health protocol. It’s subtle but important. And because of what we now know about mitochondrial health and aging, I’ve made MOTS-c part of my annual “Mito-Reset” peptide stack, alongside Epitalon and SS-31, every January.
Let’s dive into what MOTS-c is, how it works, who should consider it, and what my experience has been.
What Is MOTS-c?
MOTS-c (short for Mitochondrial Open Reading Frame of the 12S rRNA-c) is what’s called a mitochondrial-derived peptide; basically, it’s made from mitochondrial DNA instead of the usual nuclear DNA like most proteins. It was discovered in 2015 by researchers at USC, and turns out, it plays a pretty big role in things like how your body handles stress, manages metabolism, and responds to insulin. (1)
What makes this peptide unique is that it acts like a messenger: it’s produced inside the mitochondria and then travels to the nucleus to help regulate gene expression in response to cellular stress. In other words, it helps your cells adapt and survive under challenging conditions like poor diet, oxidative stress, inflammation, and aging. (2)

Benefits of MOTS-c
Although you may not feel much from MOTS-c in the short term, the science behind its longevity effect is promising. Here are some of the researched and theorized benefits:
1. Supports Mitochondrial Function
MOTS-c helps your mitochondria make energy more efficiently, especially when your body’s under stress. It supports the creation of new, healthy mitochondria and keeps the ones you have working better, longer.
2. Improves Insulin Sensitivity
Studies in mice show that MOTS-c improves glucose metabolism and increases insulin sensitivity. It might be beneficial for people with metabolic syndrome, prediabetes, or insulin resistance. (3)
3. Activates AMPK Pathway
AMPK is like the cell’s master switch for energy. When MOTS-c flips it on, it helps your body use glucose better, burn fat more efficiently, and kick off autophagy, your body’s cellular cleanup process.

Although human trials are limited, animal studies suggest that MOTS-c may increase lifespan and improve physical capacity in older subjects. It may also enhance the body’s ability to respond to exercise and environmental stress.

Kelly’s Personal Experience
When I first began researching peptides a couple of years ago, I was super curious about biohacking and longevity protocols. After trying SS-31 to repair damaged mitochondria (which I believe made a difference in my recovery and overall cellular resilience), I added MOTS-c to my protocol to build on that foundation.
Honestly? I didn’t experience a massive burst of energy or a radical transformation. But I didn’t expect to. These peptides work at a deep cellular level. The benefit is more about preventing decline, slowing aging, and building mitochondrial resilience over time.
My one complaint? The sting. Unlike most peptides I’ve injected, MOTS-c burns on injection more than anything else I’ve tried. It’s short-lived but memorable.

How to Dose This Peptide?
Most biohackers and physicians using peptide therapy recommend the following dosing protocol:
- Dosage: 5-10mg per injection
- Frequency: Daily 5 days per week on an empty stomach
- Cycle: 5 days on, 2 days off for 4 weeks; 1–2 cycles per year
- Route: Subcutaneous injection (into the abdomen, buttocks or thighs)
Some people microdose at 5mg, while others use 10–20mg per injection. Most people do not take it long-term but rather cycle it 1–2 times a year, often in conjunction with other mitochondrial peptides. Higher doses can get pricey, so that might factor into how much you decide to take.
Always consult with a peptide-literate physician or functional medicine practitioner before beginning.

Who Should Consider MOTS-c?
While MOTS-c is still in early human research stages, some groups may find it especially helpful:
- People over 40 looking to maintain mitochondrial health and delay age-related decline
- Anyone recovering from chronic fatigue, illness, or metabolic issues
- Biohackers focused on longevity, cellular health, and performance
- Athletes or older adults looking to improve endurance or recovery
Again, this peptide is not a stimulant or a “quick fix.” Its magic is in the background, optimizing how your cells handle stress and energy production.
Risks and Side Effects
Because MOTS-c is still under investigation, especially in human trials, we don’t have a complete picture of its long-term safety. However, from available data and anecdotal use, here’s what’s known:
Side Effects (Rare and Mild):
- Injection site irritation or burning (I definitely experienced this!)
- Mild fatigue or headache in some users
- Possible appetite suppression
Risks:
- Unknown long-term effects
- Not recommended during pregnancy or breastfeeding
- Should be avoided in individuals with active cancer unless under physician supervision (due to peptides influencing cell proliferation)

I’ll admit, MOTS-c didn’t blow me away with physical results. But that doesn’t mean it didn’t work. In fact, I believe the real power of peptides like MOTS-c is in what you don’t notice: the crashes you don’t experience, the illnesses you don’t get, the slow decline you don’t feel creeping in.
MOTS-c is now a staple in my annual cellular health reset protocol, and I plan to continue using it for years to come.
